摘要:
 以2001杨的春季新发嫩枝茎段、叶柄、叶片为外植体,尝试在MS和WPM基本培养基中添加多种组合激素,探索不定芽诱导、丛生芽伸长和生根最优条件,建立组织培养与植株再生系统。结果表明:MS培养基的诱导分化效果比WPM好;在MS+TDZ 0.025 mg·L-1+6-BA 0.05 mg·L-1+NAA 0.05 mg·L-1+蔗糖30.00 g·L-1的培养基上,不定芽诱导分化效果最好,茎段和叶柄诱导分化率可达90%,叶片诱导分化率 60%;丛生芽转入MS+KT 0.25 mg·L-1+GA3 1.00 mg·L-1+果糖30.00 g·L-1的培养基中茎伸长效果最佳;每个叶片外植体诱导茎伸长的芽平均达6.4个,芽数量最多;不定芽在1/2MS+IAA 0.02 mg·L-1+IBA 0.02 mg·L-1+AC 3.00 g·L-1+蔗糖30.00 g·L-1培养基上形成的根条数多,生根率达90%。
