摘要:
 以陕西省汉中市略阳县同一采收期的猪苓为研究对象,应用不同干燥方法制备样品,采用苯酚-硫酸法显色,紫外-可见分光光度法测定不同干燥方法猪苓样品的猪苓多糖含量;RP-HPLC法测定不同干燥方法制备的猪苓中麦角甾醇的含量差异;并优化猪苓药材的干燥加工方法。结果表明:紫外-分光光度法测得70℃2 h,50℃烘干法的多糖含量最高(0.668 9%),阴干法多糖含量较低;HPLC法测得自然条件晒干及50℃烘干所得麦角甾醇含量较高,而高温干燥得到的麦角甾醇含量较低。说明不同干燥方法对猪苓多糖及麦角甾醇含量有显著影响,根据不同的药用途径应当选择不同的干燥加工方法。为了提高猪苓多糖的含量,应采用先高温干燥再低温烘干加工方法,为了提高麦角甾醇含量,应采用低温烘干加工方法。
Abstract:
 Effects of different drying methods on the contents of polysaccharides (measured by UV spectrophotometer) and ergosterol (measured by HPLC) in Polyporus umbellatus were investigated.The drying procedure was optimized.The highest content of polysaccharides (0.668 9%) was found in the sample which was dried in an oven at 70℃ for 2 hours and then at 50℃ until completely dry.Ergosterol found its highest content in the samples which were undergone natural air drying and 50℃ oven drying, while lower in the samples undergone high temperature drying,indicating that drying methods significantly influenced the contents of related constituents.Suitable drying method should be adopted based on pharmaceutical applications.To obtain those with high content of polysaccharides, it was recommended the materials should be in an oven at 70℃ for 2 h,and at 50℃ until completely dry.Low temperature drying was the best choice for obtaining high ergosterol content crude drug.
