摘要:
 利用SSR分子标记,对4个白杨杂交新品种及父母本和3个近缘品种进行了指纹图谱构建和遗传关系分析。结果表明:筛选出的9对引物共扩增出106条清晰条带,平均每对引物扩增条带为11.7条,其中,多态性条带89条,多态性比率为83.1%,说明这些品种间具有较高的遗传多样性。9个品种间的遗传相似系数在0.41~0.75之间;通过系统聚类分析发现,4个杂交新品种与父本截叶毛白杨遗传相似度较高。选用扩增效果好,重复性高的3对引物构建了DNA指纹图谱,为这些品种的商业化生产应用提供了技术依据。
Abstract:
 Four new hybrids and their parents as well as 3 related varieties in Populus  were analyzed to establish their DNA-fingerprinting and the genetic relationships. Microsatellites (SSR) molecular markers were used for tree genotyping. The results showed that 106 bands were produced with 9 primer pairs selected, with a mean value of 11.7 each primer,in which the polymorphic bands were 89 with a percentage of 83.1%. It suggested that the varieties had a high level of genetic diversity. The coefficients of genetic similarity among 9 varieties varied between 0.41 to 0.75. The clustering analysis results on the basis of SSR markers revealed a high genetic similarity between 4 hybrids and their male parent P. tomentosa var. truncata. The DNA-fingerprintings constructed from 3 primer pairs with clear polymorphic patterns amplified by PCR and high reproducibility would provide the technical basis for the identification these varieties in commercial applications.
