摘要:
 以蝴蝶兰品种‘内山姑娘’的组培苗叶片为试验材料,将不同浓度的水杨酸(SA)添加于培养基中并观察叶片的褐变情况,筛选出SA的安全使用浓度为0~0.2 mmol·L-1。测定0.05、0.1和0.2 mmol·L-1的SA处理对叶片褐变指数、总酚(TP)含量、苯丙氨酸解氨酶(PAL)、多酚氧化酶(PPO)、过氧化物酶(POD)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)活性的影响。结果表明,SA处理可降低蝴蝶兰叶片褐变过程中的褐变指数、总酚含量和PAL活性,提高SOD、POD和CAT活性。0.1 mmol·L-1的SA处理抑制外植体褐变的效果较好。
Abstract:
 A Phalaenopsis cultivar ’Neyshan Guniang’ was cultured on the medium with different concentrations (0.05, 0.1 mmol·L-1 and 0.2 mmol·L-1) of salicylic acid (SA), the browning situation of the leaves cultured were observed. The browning index, the activities of TP, PAL, PPO, POD, SOD, CAT and APX were measured. The results showed that the safe dosages of SA use were between 0 to 0.2 mmol·L-1, the SA treatment could decrease browning index,total phenol content and PAL activity, and increase the SOD, POD and CAT activities. Satisfactory results of inhibiting leaf browning were achieved when the concentration of SA was 0.1 mmol·L-1.
