摘要:
 以黄精多糖为原料,研究酶解温度、酶解时间、酶解pH、酶用量对蛋白质脱除率和多糖损失率的影响。通过正交试验分析得到酶法脱除黄精多糖中蛋白质的最优工艺,并与Sevage法相比较。结果表明,酶法是最佳的脱蛋白方法,其脱蛋白最佳工艺为:木瓜蛋白酶添加量2.5%,温度60℃,时间2.5 h ,pH为6.5,蛋白质脱除率为 71.23%,多糖损失率13.7%。
Abstract:
 Enzymic hydrolysis was adopted to remove the proteins contained in the polysaccharides extracted from the rhizomes of Polygonatum sibiricum.Conditions of the hydrolysis were optimized by orthogonal experiment,and the results were compared with Sevage method.The results suggested that enzymic method was better than Sevage method.The optimal conditions in enzymic method were papain dosage 2.5%,temperature 60℃,time of hydrolysis 2.5 h,pH 6.5,under which the removal rate of deprotein was 71.23%,polysaccharide loss rate was 13.7%.
