摘要:
 对含糖型和无糖型杜仲饮料中总黄酮含量的检验方法进行了比较研究。结果表明,采用《保健食品检验与评价技术规范》(2003年版)中保健食品中总黄酮的测定方法检测杜仲饮料中的总黄酮,存在重复性提取、操作繁琐和浪费时间等缺点;进行样品水浴上挥干的前处理,对含糖型杜仲饮料中总黄酮含量的检测误差大、重现性差,检测值偏小。故不宜作为杜仲饮料总黄酮的检验方法:对检测方法进行了优化。优化后的方法精密度、重复性和加标回收率均较高,符合分析要求。优化后的杜仲饮料总黄酮的检测方法为:吸取供试样品1.0 mL于蒸发皿中,加1 g聚酰胺粉吸附,于水浴上挥去溶剂,然后转入层析柱。先用20 mL甲苯洗脱,甲苯液弃去,然后用甲醇洗脱黄酮,定容至25 mL。于波长360 nm测定吸光度,按标准曲线计算样品中总黄酮含量。用该方法检测的含糖型杜仲饮料的总黄酮含量为63.943 3 mg·100 g-1,无糖型杜仲饮料的总黄酮含量为60.322 1 mg·100 g-1。本方法可为其他液体饮料的总黄酮含量检测提供借鉴。
Abstract:
 The determination method of the contents of total flavonoids from the Technical Specification for the Determination and Evaluation of Health Care Foods (2003 edition) was adopted to determine the total flavonoids in Eucommia beverage.However,it was found that this method was not suitable to determine the total flavonoids in Eucommia beverage because it possessed some problems such as complex operation,repetitive extraction and long extraction time.Furthermore,the pretreatment of water bath evaporation showed a larger detection error,a poorer reproducibility,and a smaller detection value,which demonstrated the method should be optimized.In this work,the optimized method was determined as follows: the sample (1 mL) was mixed with polyamide powder (1 g) in an evaporating dish and evaporated with water bath,and then transferred to a chromatographic column.The chromatographic column was washed with toluene and the filtrate was discarded.Subsequently,the flavonoids were eluted with methanol and the eluate was calibrated at 25 mL.The content of total flavonoids was measured under the detection wavelength of 288 nm and calculated by the standard curve equation.According to the optimized method,the obtained total flavonoids content was 63.943 3 mg·100 g-1 in sugar beverage while 60.322 1 mg·100 g-1 in sugar-free one.It illustrated that the optimized method could not only accord with the analytical requirements for its higher precision,better repeatability and higher standard addition recovery,but also provide reference for the detection of total flavonoids content in other liquid drinks.
