摘要:
 为明确秦岭北麓猕猴桃主产区主栽品种猕猴桃溃疡病病原菌致病性强弱与地理来源及品种之间的关系,本研究在周至、杨凌和眉县选择了海沃德、华优和红阳的感病枝条为材料,采用常规组织法对猕猴桃溃疡病病原菌进行分离、纯化,结合16S-23S rDNA ITS序列分析进行分子鉴定,并进行了致病性测定。结果表明:经分离、纯化和分子鉴定,共获得丁香假单胞杆菌猕猴桃致病变种(Pseudomonas syringae pv.actinidiae)菌株20个,且不同地理来源和不同品种的菌落形态差异较大;对不同菌株致病性的测定表明,各菌株致病性强弱与其地理来源及品种之间并没有直接的关系;建立基于软件ImageJ 1.50b的定量精确叶片致病性强弱分析方法,筛选出强致病性菌株4个,可为抗性材料鉴定和抗溃疡病专用生物药剂筛选研究奠定基础。
Abstract:
 The aim of this study was to evaluate the relationships among the pathogenicity of Pseudomonas syringae pv.actinidiae (Psa) and the geographic origin and cultivars for the main cultivars in northern area of Qinling Mountains.The infected kiwifruit branches were collected from the cultivars of Hayward,Huayou and Hongyang in Zhouzhi County,Yangling District and Mei County.The pathogens of Psa were isolated and purified by the conventional tissue method.16S-23S rDNA ITS methods were used to analyze the molecular evolution of the pathogens and the pathogenicity also were measured.The results showed that:A total of twenty Psa strains were obtained by isolation,purifying and molecular identification.The colonial morphology showed a large difference for different geographic origin and cultivars.There were no relationships among pathogenicity of the strains and geographic origin and cultivars.The quantitative and accurate methods of leaf pathogenicity were established based on the software ImageJ 1.50b,and four strong strains were selected.These Psa strains provide a basis for further study on identifying resistance materials and selection of specific biological pesticide for kiwifruit bacterial canker.
