 以菊花品种‘C008’无菌苗叶片作为外植体,在建立了菊花高效再生体系的基础上,通过农杆菌介导法将CBL基因导入菊花品种‘C008’中。试验共获得抗性植株41株,PCR检测表明,阳性植株为5株,证明CBL基因已转化到菊花‘C008’的基因组DNA中,转化率为12.2%。遗传转化过程中,预培养2 d后,将OD600=0.5~0.6的农杆菌稀释50倍后侵染叶片8 min,再共培养2 d、延迟筛选2 d,有利于获得较高的转化率。
Abstract:
 On the basis of a highly efficient regeneration system of chrysanthemum, the CBL gene was introduced into chrysanthemum ‘C008’ by agrobacterium mediated method based on the chrysanthemum ‘C008’ aseptic seedling leaf as explants. In the experiment, 41 strains of resistant plants were obtained. The PCR test showed that the positive plants were 5, which proved that the CBL gene had been transformed into the genome DNA of the chrysanthemum ‘C008’, and the transformation rate was 12.2%. In the process of genetic transformation, after pre-culture for 2 d, the explants were infected for 8 min with diluted OD600=0.5-0.6 Agrobacterium culture liquild with 50 times, and then co-cultured for 2 d, delayed screening for 2 d, which was beneficial to obtain a higher transformation rate.
