摘要:
 以乌饭树(Vaccinium bracteatum)嫩枝插条为试验材料,对其取材、FAA及PFA固定液的不同固定、乙二胺及甘油乙醇溶液的不同软化处理、脱水、透明、浸蜡、包埋、切片、染色等程序进行了试验探究。结果表明,2种固定液均适宜作为乌饭树插条的固定液,乙二胺软化效果较好,染色15~20 s为宜。通过试验掌握了乌饭树茎段的石蜡切片制作方法,得出了乌饭树茎段石蜡切片制作过程中的关键技术。
Abstract:
 In the present study,the procedures for preparing paraffin section of V.bracteatum shoot cuttings were investigated.Different treatments,including drawing materials,FAA and PFA fixing solution,softening treatment with ethylenediamine and glycerol ethanol solution,dewatering,transparence,waxing,embedding,section,dyeing etc were also explored.The results showed that the two kinds of fixing solution were both suitable for the fixation of V.bracteatum stem segments.Meanwhile,softening with ethylenediamine got better effect,and the suitable dyeing time was 15-20 s.Through these experiments,the procedure of paraffin section of V.bracteatum was summarized,and the key steps in the procedure were achieved.
