摘要:
 从细叶百合(Lilium pumilum)中克隆得到LpSVP基因,该基因ORF全长为675 bp,编码224个氨基酸。序列同源性和系统进化树分析表明该基因属于MADS-box基因家族下SVP类基因;生物信息学分析表明蛋白相对分子量为25.783 kDa,理论等电点为5.62,为亲水蛋白,没有跨膜结构;亚细胞定位预测于细胞核中;蛋白质二级结构a-螺旋占60.71%,随机卷曲占29.02%,延伸链占10.27%;蛋白质三级结构以同型四聚体形式存在。半定量分析表明该基因在鳞茎、叶片、雌蕊、雄蕊和花瓣中均表达,但表达有强弱差异;实时荧光定量分析表明在花发育过程中该基因表达量先增长后下降。该研究为深入了解LpSVP基因功能奠定了基础。
Abstract:
 The LpSVP gene was cloned from Lilium pumilum,the full-length of ORF was 675 bp,and encoding 224 amino acids.Sequence homology and phylogenetic analysis showed that this gene belonged to SVP-like MADS-box genes; bioinformatics analysis showed that the relative molecular weigh of the LpSVP gene was 25.783 kDa,the theoretical isoelectric point was 5.62,and it was hydrophilic protein with no transmembrane structure; the subcellular localization was predicted in nucleus; the secondary structure analysis showed that LpSVP comprised 60.71% of alpha-helix,29.02% random coil and 10.27% extended strains; the tertiary structure of the LpSVP protein existed as isomeric tetramer.RT-PCR analysis showed that the gene was expressed in bulb,leaf,pistil,stamen and petal,but varied widely in the level of expression.Real-time PCR analysis showed that the expression of LpSVP increased firstly and then decreased continuously during flower development.The study would lay a foundation for further understanding the function of the gene.
