摘要:
 采用水提醇沉法制备紫珠草粗多糖,后经活性炭脱色、Sevage法脱除蛋白质、DEAE-52(OH-)离子交换纤维素分离纯化得到一个紫珠草多糖类物质(ZZC-PC)。对该多糖类物质进行化学测定和光谱分析,应用细胞溶血法进行抗补体活性的研究。结果表明,该物质主要含多糖与木质素,分子量为65.7 ku,具有明显的多糖特征峰;该物质具有较好的抗补体活性,其对经典途径和旁路途径的CH50为(1.95±0.1) mg·mL-1、AP50为(3.3±0.2) mg·mL-1
Abstract:
 Crude polysaccharide was obtained from Callicarpa macrophylla by water extraction and ethanol precipitation method.Then the crude polysaccharides were purified by active carbon decoloration,sevage deproteinization and DEAE-52 (OH-) ion exchange cellulose to prepare the polysaccharide (ZZC-PC).The polysaccharide was characterized by chemical and spectral analysis and its anti-complementary activity was evaluated by the hemolysis assay.The results showed that ZZC-PC was mainly composed of polysaccharides and lignin possessing obvious absorption peaks of polysaccharide,and the molecular weight (MW) was 65.7 ku.It had potent anti-complementary activity,which inhibited complement activation on both the classic and alternative pathways with CH50 value of (1.95±0.1) mg·mL-1 and AP50 value of (3.3±0.2) mg·mL-1,respectively.
