摘要:
 为优选出大岩桐组织培养中最佳的消毒灭菌方法,以大岩桐幼嫩叶片为试验材料,研究前期75%酒精(10、30、60 s)预处理以及后期0.1% HgCl2、5% NaClO、10% H2O2 3种不同消毒剂种类分别在(8、10、12 min)处理下对大岩桐生长状况的影响。结果表明,10% H2O2处理下的长菌率和褐化率显著高于0.1% HgCl2和5% NaClO的处理,且长菌和褐化发生时间较早;研究中消毒效果最佳的2个组合为75%酒精处理30 s+0.1% HgCl2处理10 min和75%酒精处理30 s+5% NaClO处理10 min。该研究为与大岩桐类似的其他被子植物的组织培养提供了参考。
Abstract:
 In order to optimize the sterilization method in tissue culture of Sinningia speciosa,young leaves of S.speciosa were used as experimental materials.The effects of pretreatment with 75% alcohol (10,30,and 60 s) in the early stage and post-treatment with 0.1% HgCl2,5% NaClO and 10% H2O2 (8,10,and 12 min each) on the growth of S.speciosa leaves were studied.The results showed that the pollution rate and browning rate under 10% H2O2 treatment were significantly higher than those of 0.1% HgCl2 and 5% NaClO,and the pollution and browning occurred earlier.The two most effective combinations were 75% alcohol treatment for 30 s+0.1% HgCl2 for 10 min and 75% alcohol treatment for 30 s+5% NaClO for 10 min.This experiment provides a reference for tissue culture of other angiosperms similar to globulus.
