摘要:
 为了研究白榆芽变突变体中白化叶片的光合特性以及光合通路中突变基因转录和翻译水平表达调控机制,以培育3 a的白榆芽变突变体无性系植株上的白化以及正常绿色叶片为试验材料,对其进行光合色素含量测定、叶绿素荧光参数测定以及无参转录组测序分析。结果表明,白化叶片中的叶绿素a(Chla)、叶绿素b(Chlb)、类胡萝卜素(Caro)和总叶绿素(Chla+b)均显著低于正常叶片,叶绿素a/b(Chla/Chlb)与类胡萝卜素与总叶绿素含量(Ccaro/Chl)的比值也显著降低(P<0.05);白化叶片的Fo、Fm、Fv/Fo、Fv/Fm、ΦPSⅡ、qP和NPQ等参数均与正常叶片出现显著差异(P<0.05),且白化叶片Fv/Fm值为0.619;而NPQ值出现白化叶片高于正常叶片,说明白化叶片在正常环境中,明显出现某些伤害。利用转录组测序技术,共获得14.38 Gb有效数据(CleanData),对Unigene进行功能注释,共获得9 226条Unigene的注释结果;对Unigenes表达量分析,共获得2 923条差异基因(DEGs),在白化叶片有1 250个基因表达为上调,1 673个基因表达为下调;对注释信息进行KEGG富集分析得到,以卟啉与叶绿素代谢过程中相关蛋白的编码基因、编码光系统Ⅱ、Ⅰ结构蛋白的psb、psa和pet以及编码光合作用-天线蛋白的LHC基因家族的下调表达,推测与白榆芽变突变体出现光合色素含量以及光系统性能降低现象有关。
Abstract:
 In order to study the photosynthetic characteristics of the albino leaf in the bud mutant of Ulmus pumila and the regulation mechanism of the transcription and translation level of the mutant gene in the photosynthetic pathway,this paper took the albino and normal leaves on the clones of the bud mutant of U.pumila cultivated for 3 years as experimental materials.The photosynthetic pigment content determination,chlorophyll fluorescence parameter determination and transcriptome-free sequencing analysis were performed.The results showed that the chlorophyll a (Chla),chlorophyll b (Chlb),carotenoid (Caro) and total chlorophyll (Chla+b) in the albino leaves were significantly lower than those in the normal leaves,and the contents of Chla/Chlb,the ratio of total carotenoid to chlorophyll content (Ccaro/Chl) also decreased significantly (P<0.05).Parameters of Fo,Fm,Fv/Fo,Fv/Fm,ΦPSⅡ,qP and NPQ of albino leaves were also significantly different from normal leaves (P<0.05),and the Fv/Fm value of the albino leaves was 0.619; while NPQ values of the albino leaves were higher than those of normal leaves,indicating that some damages occurred in the albino leaves under normal environment.Using transcriptome sequencing technology,a total of 14.38 Gb of effective data (CleanData) was obtained.A total of 9 226 Unigene annotation results were obtained from functional annotation.For the analysis of Unigenes expression,a total of 2 923 differential genes (DEGs) were found in the albino leaves,in which 1 250 genes were up-regulated and 1 673 genes were down-regulated.KEGG enrichment analysis was performed on the annotated information.The genes encoding the related proteins in the metabolism of porphyrin and chlorophyll,the psb encoding the structural proteins of photosystem II and I,the down-regulated expression of psa and pet,and the LHC gene family encoding the photosynthesis-antenna protein were presumed to be related to the appearance of photosynthetic pigment content in the bud mutant of U.pumila and a decrease in the performance of the photosystem.
