摘要:
 以杨树叶片为试验材料,采用改良SDS 法提取基因组DNA,分析5种不同保存方法对样品提取效果的影响。通过A260 /A280、琼脂糖凝胶电泳、ISSR-PCR扩增分析对提取的基因组DNA进行鉴定,结果表明,低温保存杨树叶片是比较理想的方法, 而在远距离野外资源调查收集时,采用硅胶干燥保存样品是比较可行的。
Abstract:
 Effects of different preservation methods on the extraction of genomic DNA with improved SDS method from poplar leaves were studied. The extracted DNA was tested by using the value of A260 /A280, the agarose gelelectrophoresis, and ISSR-PCR amplification. The results showed that low temperature preservation of poplar leaves was the best method. However, silica gel preservation was a feasible alternative for preserving the samples collected the field resources investigation.
