剪取四倍体刺槐当年生嫩枝作插穗,研究了不同浓度和时间外源激素IBA浸泡处理后对嫩枝插穗生根的影响及生根过程中内源激素IAA、腺嘌呤、ABA、GA 、IBA的变化,以揭示四倍体刺槐插穗难生根的机理。结果表明:1)在扦插生根过程中,插穗体内IAA、腺嘌呤、GA含量在0~15 d时上升,15~30 d时下降; 不定根形成前需要较高IAA水平,以诱导生根,不定根形成后,IAA合成减弱,以保持正常生长。2)插穗腺嘌呤含量与嫩枝插穗愈伤组织诱导率无显著相关性,但在不定根形成前(0~15 d)需要一定量的腺嘌呤以促进愈伤组织诱导和分化。3)插穗GA含量与嫩枝插穗愈伤组织诱导率有显著负相关性,不定根形成前GA含量的上升很可能是抑制不定根形成的主要原因。4)插穗ABA和IBA含量与嫩枝插穗愈伤组织诱导率无显著相关性。
Abstract:
In order to understand the causes of the difficulty in rooting, current year soft wood cuttings of tetraploid locust were treated by an exogenous hormone, IBA with different concentrations and time of treatment to examine their effects on rooting and the levels of endogenous hormones, such as IAA, adenine, ABA, GA, and IBA. The results showed that: 1)During the rooting process, contents of IAA, adenine and GA in the cuttings increased in the first 0-15 days, but decreased during 15-30 days. Adventitious roots formation required high IAA level, and then IAA synthesis weakened in order to maintain the normal growth. 2)No significant correlation was observed between adenine content and callus induction rate, certain amount of adenine was needed to promote callus induction and differentiation before the formation of adventitious root(0-15 d). 3)Significantly negative correlation was found between GA content and callus induction rate, and the increase of the levels of GA before adventitious root formation was likely to be the main reason for inhibition of adventitious root formation. 4)ABA and IBA contents in softwood cuttings had no significant correlation with callus induction rate.
