摘要:
 以葡萄无核品种无核白和有核品种黑比诺为材料,克隆葡萄CONSTANS基因家族的1个成员并将其命名为VvCOL5基因,分析基因结构,进行染色体定位和系统发育树的构建,检测VvCOL5基因在无核和有核葡萄胚珠(幼种子)发育过程的表达变化。结果表明:VvCOL5基因cDNA长度为1 374 bp,包含有2个外显子和1个内含子结构,并含有2个B-box结构域和1个CCT结构域,属于CO家族的第一类,在GenBank登录号为KX077245。在受精后胚珠(幼种子)发育过程中,VvCOL5基因在无核白与黑比诺中呈现出了相反的表达模式。其无核白受精后10~30 d其表达缓慢上调,30~35 d维持在较高水平,35~40 d缓慢下降,后在40~45 d又快速上调到较高水平;而在黑比诺中10~20 d下降,在20~35 d之间相对平稳,35~40 d缓慢上升,最后40~45 d时又下降到较低水平。无核白中VvCOL5基因在各时期的表达量均高于黑比诺,可见VvCOL5基因在无核和有核品种中表达差异较大,其与葡萄种子发育有一定关系。
Abstract:
 One CONSTANS gene was cloned from seedless (cv.Thompson seedless) and seed (cv.Pinot Noir) cultivars.Gene structure analysis,chromosome,mapping analysis,and phylogenetic analysis were performed.The expression patterns of VvCOL5 gene at different stages of ovule development in nuclear grapes and seedless grapes were investigated.VvCOL5 gene contained two extrons and one intron with cDNA sequence length of 1 374 bp.It contained twoB-box domains and one CCT domain,belonging to the first type in CO family gene.The expression patterns of VvCOL5 gene were opposite in ovule development between cv.Pinot Noir and cv.Thompson seedless cultivars.The ovule expression in cv.Thompson seedless gradually up-regulated at 10-30 DAF,maintained higher level at 30-35 DAF,gently down-regulated at 35-40 DAF and quickly up-regulated to the high level at 40-45 DAF.However,Pinot Noir showed expression that down-regulated at 10-20 DAF (day after full-bloom),relatively stable quantities at 20-35 DAF,slowly up-regulated at 35-40 DAF and down-regulated to the lower level at 40-45 DAF.In addition,the VvCOL5 expression quantities in cv.Thompson seedless was higher than cv.Pinot Noir in the whole period.This research showed that VvCOL5 gene existed significant difference between seedless and seed varieties,which may related to grape seedlessness.
