摘要:
 采用傅里叶变换红外光谱、二阶导数谱和二维相关红外光谱技术对不同干燥温度的黄芩药材进行分析研究。结果表明,不同干燥温度的黄芩药材一维光谱非常相似,相关系数随着温度的升高逐渐加大。黄酮类成分1 614 cm-1附近特征吸收峰及糖类成分1 055 cm-1附近吸收峰的相对峰强度差别显著。不同干燥温度的黄芩药材二维相关红外光谱存在差异,在糖环振动区域800~1 000 cm-1波段内的二维相关谱比较,黄芩采用80℃、100℃和120℃干燥均出现了2个强自动吸收峰,在糖骨架振动区域1 000~1 200 cm-1,黄芩采用自然干燥和60℃干燥,有3个强自动吸收峰,最强吸收峰在1 094 cm-1;采用80℃、100℃和120℃干燥,3个样品自动吸收峰强度发生变化,最强吸收峰在1 005 cm-1。表明红外光谱法结合二维相关红外光谱技术可为同一批次药材采用不同的干燥温度所存在的细微差别分析与研究提供一种快速方法和手段。
Abstract:
 Fourier transform infrared spectroscopy (FTIR) combined with two-dimensional correlation spectroscopy (2D-IR) was used to analyze and evaluate Scutellaria baicalensis roots that were dried at different temperatures.The results showed that single-dimensional FTIR of the roots dried at different temperatures were very similar,and the correlation coefficient increased gradually with the increase of temperature.The relative intensities of the characteristic absorption peaks at 1 614 cm-1 for flavonoids and at 1 055 cm-1 for glycosyl were different.However,There were differences in the spectra of 2D-IR among the roots dried at different temperatures.Comparing the 2D-IR spectra:in the band of 800-1 000 cm-1 (due to the vibration of glycosyl),two strong peaks were observed for the roots dried at the temperatures of 80℃,100℃,and 120℃,while in the band of 1 000-1 200 cm-1,three strong peaks were found for the air-dried roots and the roots dried at 60℃,and the strongest one was 1 094 cm-1.The peak with the strongest intensity for the roots dried at 80℃,100℃ and 120℃ was at 1 005 cm-1.Therefore,FTIR combined with 2D-IR was fast and comprehensive approach to analyze and evaluate the imperceptible differences among the roots dried at different temperatures in the same batch of Chinese medicinal materials.
