摘要:
 以经外源IBA、NAA和ABT1处理后的国槐插穗为材料,应用酶联免疫吸附分析法(ELISA),研究其在嫩枝扦插生根过程中4种内源激素的变化。结果表明:与对照相比,生根促进剂显著提高了插穗生根率,不同生根促进剂对插穗生根作用显著,尤其以IBA处理插穗的生根表现最好,其生根率达35.17%,显著高于NAA(24%)、ABT1(17%)处理。在生根促进剂处理下,4种内源激素IAA、ABA、GA3、ZR在插穗不定根形成过程中含量发生相应变化,进而影响插穗生根。在不定根扦插初期,即生根促进剂处理后12 d左右,与对照相比,各处理插穗的IAA含量、IAA/ABA和IAA/ZR比值处于较高水平,ABA、GA3、ZR含量处于较低水平。生根率和生根时间接近的NAA处理和ABT1处理的内源激素变化规律相似。IBA处理显著提高了不定根诱导期IAA水平,但使ABA、GA3、ZR的含量显著降低,并促使IAA含量顶峰提前到来,这是IBA处理后插穗生根效果显著提高的重要原因。
Abstract:
 Cuttings of Sophora japonica treated with IBA,NAA and ABT1 were used as materials to investigate the changes of four endogenous hormones in cuttings during the rooting process by means of ELISA.The results showed that the application of rooting accelerators changed the contents of four endogenous hormones (IAA,ABA,GA3 and ZR) in softwood cuttings of S.japonica in the process of adventitious root formation.The significant differences were found among three treatments.The rooting accelerators treatments increased rooting rate significantly.The rooting rate of the cuttings treated by IBA was 35.17%,which was higher than that treated by NAA (24%) and ABT1 (17%).About 12 days after cutting,the endogenous IAA content and the IAA/ABA,IAA/ZR ratio was at relatively high levels,and the endogenous ABA,GA3 and ZR were relatively low.The fluctuations of endogenous hormones were similar between NAA and ABT1 treatment for their similar rooting rates and similar rooting time.IBA increased the content of IAA and promoted the earlier coming of climaxes of IAA,while it could decrease the contents of ABA,GA3 and ZR,which were accounted for the improvement of rooting ability after treated with IBA.
