摘要:
 前期研究表明,拟南芥初生壁纤维素合成酶(CESA)的磷酸化修饰是调控纤维素合成和细胞形态建成的重要机制。以拟南芥CESA为对照,对杨树、葡萄、番茄、苜蓿以及小麦、大麦、玉米、水稻和大豆等13个物种的CESA家族成员进行序列比对和系统发育分析,结合相关磷酸化蛋白组学数据的收集和整理,分析鉴定纤维素合成酶磷酸化位点的保守性。蛋白组学数据分析表明,拟南芥和作物CESA中已被质谱技术鉴定出的磷酸化位点主要集中在植物特有的3个区域,包括2个序列高变的CESA种类特征区。系统发育树分析表明,CESA蛋白家族聚为7大类,包括初生壁的CESA1,3和6家族,次生壁的CESA4,7和8家族,以及苔藓和石松CESA家族。其中,初生壁CESA1和CESA3家族的一些磷酸化位点在苔藓和石松CESA家族中高度保守,表明CESA的磷酸化修饰可能是初生壁纤维素合成的一种相对保守的分子调控机制。分析结果进一步表明,次生壁CESA也含有多个磷酸化位点。其中,CESA4家族的一些磷酸化位点在进化中也具有高度保守性,首度表明,除了转录水平调控外,次生壁CESA的磷酸化修饰可能同时是次生壁纤维素合成的重要分子调控机制。
Abstract:
 Our previous study demonstrated that phosphorylation of primary wall cellulose synthases (CESAs) plays a crucial role in regulating primary wall cellulose biosynthesis and plant cell morphogenesis.In this paper,we investigated the evolution of CESA phosphorylation sites to understand their conservation and divergence across plant species.Sequence and phosphoproteomics data of cellulose synthases were downloaded and organized from 13 species,including poplar,Medicago,wheat,barley,maize,rice,soybean,tomato,and grape.Multiple phosphorylated residues were identified in primary wall CESAs,which wre mainly clustered in three domains of CESAs,including two highly variable class-specific domains.Some phosphorylation sites of primary wall CESAs,including CESA1 and CESA3,were conserved across the CESA clades from moss and lycophyte,suggesting that regulation of primary cell wall biogenesis through CESA phosphorylation is a relatively ancient event.Multiple phosphorylation sites were also identified in CESAs for secondary wall biogenesis,and some of the phosphorylation sites were conserved across the CESA clades from moss and lycophyte as well,suggesting that in addition to regulation at transcriptional levels,CESA phosphorylation may also play important roles in regulating cellulose biosynthesis in the secondary wall.
