摘要:
 以枣雄性不育种质‘JMS2’为母本、酸枣优系‘邢16’为父本通过人工控制杂交获得的179株杂交后代及其父母本为试验材料,利用SSR标记进行杂种鉴定及遗传多样性分析。结果表明,利用双亲差异互补SSR引物JSSR311一次性鉴定出所有子代均为杂种,实现了该组合大量杂交后代的快速、高效鉴定。随机选取21对SSR引物,在杂交后代中共扩增出46个等位基因位点,其中多态性条带24个,多态性比率52.17%。经遗传分析,杂交后代群体Shannons信息指数(I)平均值为0.64,期望杂合度(Hei)平均值为0.42,说明杂交后代群体的遗传多样性高。UPGMA聚类分析显示,杂交后代个体及亲本间相似系数介于0.64~0.97,在相似系数0.70附近可将杂交后代分为3大类,其中第1大类由172个子代与母本聚在一起、占子代总数的96%,第2大类由2个子代与父本单独聚为一类,表明杂交后代与母本JMS2的遗传相似度较高。
Abstract:
 The male sterile Chinese jujube (Ziziphus jujuba Mill.‘JMS2’) and wild jujube (Z.acidojujuba Cheng et Liu ‘Xing 16’),as well as their 179 hybrid progenies were used.SSR markers were adopted to identify the hybrids and to conduct the genetic diversity analysis.The results showed that the JSSR311,a two-parent differential primer,could quickly identify the hybrid purity of mass progeny and confirmed that all the tested progenies were hybrids.A total of 46 bands/gene loci were amplified from 21 randomly selected SSR primers,of which 24 bands were polymorphic (52.7%).The Shannons information index (I) was 0.64 and the expected heterozygosity (Hei) was 0.42,which showed a high genetic diversity in hybrid progeny.The UPGMA dendrogram demonstrated that the genetic similarity coefficient between individuals and parents ranged from 0.64 to 0.97,the hybrids could be divided into three classes at the similarity coefficient of 0.70.The first category included 172 progenies and female parent ‘JMS2’,accounting for 96% of the total number of offspring.The second category included 2 progenies and male parent wild jujube,indicating that the hybrid progeny had higher genetic similarity with the female JMS2.
