摘要:
 以‘朝霞’楸带腋芽的茎段为试验材料,通过正交试验设计,研究基本培养基、植物生长调节剂成分及含量对不定芽增殖及生根的影响。结果表明,继代培养阶段,基本培养基类型是影响增殖系数的主要因素,不定芽增殖最佳培养基为:DKW+6-BA1.6 mg·L-1+NAA0.01 mg·L-1,增殖系数高达3.1。生根培养阶段,经极差分析和方差分析得知生根率最高的培养基为WPM+NAA0.1 mg·L-1,生根率高达97%,添加物活性炭对根数的增加有抑制作用,‘朝霞’楸最适生根培养基为WPM+NAA0.1 mg·L-1+活性炭0 g·L-1。本研究旨在筛选楸树优良品种‘朝霞’楸不定芽增殖和生根培养基,为建立‘朝霞’楸再生体系提供技术支持。
Abstract:
 Catalpa bungei is an important timber and ornamental tree species in China.It is of significance to study the key techniques of tissue culture procedure for improved variety of this species.The aims of this study were to select the optimum media of rooting and the multiplication of adventitious buds in tissue culture for C.bungei ‘Zhaoxia’,and to provide technical supports for establishing tissue culture regeneration system of this improved variety.Taking stems with axillary buds of C.bungei ‘Zhaoxia’ as testing materials,effects of different basic media,different plant growth regulators and their concentrations on adventitious bud multiplication and rooting were studied by using orthogonal experiment design.The results showed that the culture medium was the main factor influencing the proliferation coefficient in the subculture stage,the optimum medium for adventitious bud multiplication was DKW+6-BA1.6 mg·L-1+NAA0.01 mg·L-1,by which the average proliferation coefficient was 3.1.The orthogonal experiment and ANOVA showed that the optimum medium for rooting rate was WPM+NAA0.1 mg·L-1,by which the rooting rate was 97%.In addition,activated carbon had an inhibitory effect on increasing root number,the optimal rooting medium was WPM+NAA 0.1 mg·L-1+activated carbon 0 g·L-1.
