摘要:
 以华中五味子藤茎为材料,采用硅胶柱层析、MCI树脂柱层析、Sephadex LH-20凝胶柱层析等方法,分离鉴定所得化合物的结构,采用ABTS方法评价其抗氧化活性,以全面了解华中五味子的化学成分,明确其藤茎中药效成分及活性作用。结果表明:1)从藤茎乙酸乙酯萃取部位中分离得到7个化合物,分别鉴定为胡萝卜苷(1)、pinobatol (2)、Matairesinol 4′-O-glucoside (3)、(-)-secoisolariciresinol-O-a-L-rhamnopyranoside (4)、(-)-secoisolariciresinol 9-O-α-L-arabinopyranoside (5)、(-)-儿茶素 (6) 和异鼠李素-3-O-β-D-葡萄糖苷(7);2)化合物2~5为首次从该属植物中分离得到;3)化合物2~7清除ABTS自由基IC50值分别为14.43、13.45、7.39、3.84、1.88、14.11 μg·mL-1;化合物4~6抗氧化活性与阳性对照品相当。
Abstract:
 Taking the canes of Schisandra sphenanthera as materials,chemical constituents were thoroughly investigated and their antioxidant activities were evaluated in order to explicit active constituents and action mode.The chemical constituents were isolated and purified by various chromatographic techniques,such as silica column chromatography,MCI resin column chromatographic and Sephadex LH-20 column chromatographic methods.Their structures were elucidated on the basis of spectroscopic methods,and the antioxidant activities of compounds 2-7 were evaluated by ABTS free radical scavenging assay.The results were as follows:1) seven compounds were elucidated on the basis of spectral and chemical evidences,i.e.,daucosterol (1),pinobatol (2),Matairesinol 4′-O-glucoside (3),(-)-secoisolariciresinol-O-α-L-rhamnopyranoside (4),(-)-secoisolariciresinol 9-O-α-L-arabinopyranoside (5),(-)-catechin (6),and isorhamnetin-3-O-β-D-glucoside (7).2) Compounds 2-5 were isolated from Schisandra for the first time.3) Compounds 2-7 presented significant scavenging activities against ABTS free radicals,the IC50 values of which were 14.43,13.45,2.93,7.39,3.84,1.88 μg·mL-1 and 14.11 μg·mL-1,respectively.Compounds 4-6 exhibited similar antioxidant activities to positive control.
