摘要:
 为分析白杨派优良无性系之间的遗传关系,采用毛细管电泳荧光SSR分子标记技术,构建10个白杨派优良无性系DNA指纹图谱,并进行遗传分析。结果表明,从20对引物中筛选出7对位点多态性高且重复性好的引物,共扩增出41个多态性位点,平均每对引物扩增5.9个,每个引物扩增多态位点在4~8之间。无性系间遗传相似系数区间为0.415~0.902,平均遗传相似系数为0.657,遗传变异性丰富。Shannon指数和基因多样性指数分别为1.433 4和0.694 3,表明无性系间具有较高的遗传多样性。聚类分析表明,10个白杨派无性系中,秦白杨1号、秦白杨3号、秦白杨5号与84K聚为一类,07-17-18、07-23-23与07-30-11与I-101聚为一类,与实际亲缘关系相符。利用2对多态性SSR引物PMGC-2020和PMGC2607构建了白杨派无性系指纹图谱,该图谱可将无性系全部区分开。
Abstract:
 To analyze the genetic relationships of poplar clones (Sect.Leuce),we constructed DNA fingerprint maps of 10 excellent poplar clones and analyzed genetic diversity based on fluorescent SSR molecular markers of capillary electrophoresis.A total of 41 polymorphic loci were detected in the 7 pairs of polymorphic primers with high polymorphism and high repeatability,20 pairs of primers were screened out.The polymorphic loci of each primer ranged from 4 to 8 with an average of 5.9.The genetic similarity coefficient ranged from 0.415 to 0.902,with the average of 0.657.The Shannon index and gene diversity index were 1.433 4 and 0.694 3,respectively,indicating that there was a high genetic diversity.Cluster analysis showed that “Qinbaiyang No.1”,“Qinbaiyang No.3”,“Qinbaiyang No.5” and “84K” were clustered in one group,and “07-17-18”,“07-23-23”,“07-30-11” and “I-101” were clustered another group,which was consistent with the actual genetic relationship.All clones could be distinguished by the fingerprints built by the primers of PMGC-2020 and PMGC2607.
