摘要:
 为了解甘肃黄河沿岸枣品种的遗传多样性,用SSR分子标记对39份枣品种进行遗传分析。20对SSR引物共扩增出142个条带,多态性条带百分率达100%。每对引物扩增的条带数为3~13个,平均每对引物扩增的条带数为7.10个,扩增片段大小为70~300 bp。引物BFU0308扩增的条带数最多,为13条,扩增条带最少的引物为JSSR198和JSSR289,各有3条。用NTSYS-pc V2.10软件计算的39份枣品种的遗传距离(GS)为0.000 0~0.993 8,平均遗传距离为0.558 7。结果表明,39份枣品种间遗传差异较大,遗传多样性丰富。UPGMA聚类图将39份枣品种分为两大类群Ⅰ和Ⅱ,第Ⅱ类群又分为4个亚群。结果表明,枣品种间的亲缘关系与品种原产地有一定相关性。亲缘关系近的品种有晋矮4号和晋矮3号、鸣山大枣、敦煌大枣、小口枣和开口酥等。SSR聚类分析结果也证实“大王枣”是与晋矮4号、晋矮3号起源相同的品种,“冷白玉2”是芒果冬枣类品种,“晋赞大枣”是壶瓶枣的同源品种。SSR分子标记技术揭示了甘肃黄河沿岸枣品种具有丰富的遗传多样性和复杂的亲缘关系,为有效解决枣品种命名混乱问题提供了依据,也为进一步研究和利用这些枣种质资源提供了依据。
Abstract:
 In order to understand the genetic diversity for jujube cultivars planted in different areas along the Yellow River in Gansu Province,a total of 39 jujube cultivars were selected and analyzed based on 20 SSR markers.The genetic relationships among the cultivars were determined.A total of 142 bands were amplified from the cultivars selected,with 100% of polymorphic bands.There were 3 to 13 bands from each marker,with an average of 7.10 bands per marker,and the amplified fragment size ranged from 70 to 300 bp.The marker BFU0308 obtained the most bands (13),while only 3 bands were obtained for the markers JSSR198 and JSSR289.The values of genetic distance (GS) among 39 jujube cultivars were calculated by NTSYS-PC V2.10 software,which were from 0.000 0 to 0.993 8,with an average of 0.558 7.The results showed significant genetic differences and rich genetic diversity among the 39 jujube cultivars.The 39 cultivars were clustered into two groups (Ⅰ and Ⅱ) by UPGMA method,and group II could be further divided into four subgroups.The clustering results were basically consistent with the cultivars’ origins.Close relationships were found among some cultivars,such as "Jinai No.4" and "Jinai No.3","Mingshandazao","Dunhuangdazao","Xiaokouzao",and "Kaikousu".The results of SSR cluster analysis also confirmed that "Dawangzao" originated from the same variety as "Jinai No.4" and "Jinai No.3","Lengbaiyu No.2" was mango winter jujube,and "Jinzandazao" was a homologous cultivar of "Hupingzao" with a high close relationship.It was concluded that the SSR marker technology revealed the rich genetic diversity and complex genetic relationships among jujube cultivars planted along the Yellow River in Gansu Province,which could provide useful information for solving the confusion in the naming of jujube cultivars,and also provided the basis for further research and utilization of these germplasm resources.
