摘要:
 将瑞香狼毒内生真菌孢子悬液接种于以瑞香狼毒提取物为主要碳源的PDA培养基中,分别在培养的第0、3、5、7、9、12 天时,采用可见紫外分光光度法测定体系中黄酮和香豆素类成分含量,考察不同接种量(10%、20%、30%)、初始pH(5、6、7、8、9、10)和外加碳源(葡萄糖0、0.2%、0.5%、1.0%、1.5%、3.0%)对降解性能的影响。在培养第3天时,瑞香狼毒内生真菌C1Y74株对黄酮和香豆素类成分的降解作用明显,培养体系中残留量分别为432.24 mg/L和1.25 mg/L,极显著低于对照组474.68 mg/L和1.61 mg/L(p<0.01),在第12 天时达最低值,分别为303.78 mg/L和0.43 mg/L。在孢子接种量30%(V/V)、添加葡萄糖1%、初始pH=5时,体系中黄酮类化合物残留量为145.08 mg/L,极显著低于对照组568.27 mg/L(p<0.01),不添加葡萄糖时,体系中香豆素类化合物残留量为0.08 mg/L,极显著低于对照组0.61 mg/L(p<0.01)。
Abstract:
 The spore suspension of the endophytic fungi was incubated to the PDA culture medium with the addition of the extract of Stellera chamaejiasme as the main carbon source.The contents of residual flavonoids and coumarins in the cultural system were determined by ultraviolet-visible spectrophotometer,and the degradation effects of the initial inoculum size (10%,20%,30%), initial pH (5,6,7,8,9,10) and additional carbon source (glucose,0,0.2%,0.5%,1.0%,1.5%,3.0%) were investigated.It was found that at the 3rd day of the incubation,degradations of flavonoids and coumarins were obvious,and their contents in the residues were 432.24 mg/L and 1.25 mg/L,respectively,significantly lower than those in the control group: 474.68 mg/L and 1.61 mg/L (p<0.01).At the 12th day,the corresponding values were 303.78 mg/L and 0.43 mg/L,respectively.When the inoculating amount of spora was 30%(V/V),the adding glucose was 1% and the initial pH was 5,the content of residual flavonoids in the system was 145.08 mg/L,significantly lower than that of the control group 568.27 mg/L,(p<0.01).When no adding glucose,the content of residual coumarins in the system was 0.08 mg/L,significantly lower than that of the control group 0.61 mg/L(p<0.01).
