摘要:
 为建立一套快速、有效的康乃馨白色品种的再生体系,以康乃馨‘白雪公主’的幼叶为外植体,对其愈伤组织和不定芽诱导、不定芽伸长和不定根诱导各阶段所需的植物激素、植物材料的影响进行了探讨。结果表明,当叶片外植体的长度为5 mm时,在TDZ 0.5 mg·L-1、NAA 0.5 mg·L-1的MS培养基上,愈伤组织和不定芽的诱导效果最为理想;从愈伤组织切下不定芽,移植于单独添加NAA为0.5 mg·L-1的培养基能有效地促进不定芽的伸长;不定芽伸长至3~4 cm后,在添加NAA 0.1 mg·L-1的培养基上,不定根分化的数量最多;再生植株经移栽和炼苗后生长状态良好。
Abstract:
 In order to develop a rapid and efficient method for the regeneration of white carnation (Dianthus caryophyllus) cultivar, the leaf cuts from White Diana were used as explants, and the effects of plant hormones and materials on the stages of callus and adventitious bud inducement, shoot elongation, adventitious root inducement were determined. The results showed that the most calli and adventitious buds were induced from the leaf explants in the length of 5 mm on the MS basal medium supplied with 0.5 mg·L-1 TDZ combined with 0.5 mg·L-1 NAA. The adventitious buds which had been excised from the residual calli could be facilitatively elongated to shoots by transferring them to the medium with 0.5 mg·L-1 NAA alone. Shoots longer than 3~4 cm cultured on the medium supplied with 0.1 mg·L-1 NAA produced a maximum number of roots per shoot. Regenerated plantlets could be successfully acclimatized and transplanted to pods.
