摘要:
 花粉超低温保存后花粉活力因材料不同有很大差异,导致这种差异的原因尚不完全清楚。以液氮保存前后花粉萌发率变化不同的2个芍药品种为对象,研究了花粉在超低温保存前后活性氧及其膜脂过氧化的关系,以探讨超低温保存对它们的影响。结果表明,2个品种保存前后萌发率变化不同,但活性氧水平均无显著变化;2个品种的抗氧化酶活性均上升,抗坏血酸含量在冻存后也明显上升,而‘大红赤金’冻存前后无显著变化;同时,液氮保存前后‘大红赤金’的丙二醛含量和相对电导率均明显上升,而‘紫凤朝阳’无显著变化。以上表明,虽然这2个品种的活性氧水平均没有显著差异,但抗氧化系统已启动并清除了多余的活性氧,超低温保存对花粉的生理有一定影响。其中花粉生活力下降的‘大红赤金’发生了膜脂过氧且抗坏血酸含量无显著变化,表明抗坏血酸的作用可能更关键。
Abstract:
 Different plants exhibit quite different pollen viabilities after cryopreservation. However, the mechanism is not completely clear yet. Taken 2 peony varieties (Dahongchijin and Zifengchaoyang) whose pollen germination rate showed different changes after liquid N2 storage as materials, this research investigated reactive oxygen species (ROS) and lipid peroxidation (LPO) of the pollen before and after the storage, and then discussed the influence of cryopreservation on ROS and LPO. The results showed the ROS content didn’t change significantly,although the pollen germination rate of 2 varieties showed different changes. The oxidative enzyme activity of both increased, but the increase of ‘Zifengchaoyang’ whose germination rate kept the same after cryopreservation was obviously higher than ‘Dahongchijin’ whose germination rate decreased. The ascorbic acid (ASA) content of ‘Zifengchaoyang’ increased dramatically after LN2 storage, while that of ‘Dahongchijin’ did not. The MDA content and relative electrolytic leakage both rose in ‘Dahongchijin’, but kept the same in ‘Zifengchaoyang’. As a conclusion, the 2 varieties showed no significant differences in ROS content, but the antioxidative system had been activated and cleared the redundant ROS. Cryopreservation had influences on pollen physiology. The variety whose pollen germination rate had decreased, ‘Dahongchijin’, showed the happening of lipid peroxidation and ASA content didn’t go up. It indicated that ASA may play a crucial role.
