摘要:
LAZY1基因对于植物分枝角度、分枝方向具有重要影响。以沙柳为试验材料克隆LAZY1基因,利用生物信息学软件分析LAZY1基因序列、构建进化树,并预测蛋白结构。结果表明,SpsLAZY1aSpsLAZY1b的CDS全长分别为1 161、1 143 bp,分别编码386和380个氨基酸。生物信息学分析表明SpsLAZY1a、SpsLAZY1b 2种蛋白均为亲水性蛋白,均无信号肽区域。氨基酸序列具有IGT基因家族典型结构域,并且其C端有LAZY1蛋白的典型EAR结构。系统发育树将18个LAZY1蛋白家族聚为2个大类, SpsLAZY1a和SpsLAZY1b属于其中第1类,均为双子叶植物,并与其他植物具有较高相似性。氨基酸同源结构比对发现这些植物间既有相似同源区域,也存在一定差异区域。单子叶比双子叶植物缺少某些同源区域,草本比木本植物又缺少某些同源区域。LAZY1基因在沙柳进化过程中发生基因重复,本研究由沙柳中成功克隆SpsLAZY1aSpsLAZY1b基因,隶属IGT基因家族,并初步揭示LAZY基因的系统进化关系。
Abstract:
 The LAZY1 genes have important influence on the branching angle and direction in plants.SpsLAZY1a and b are cloned from Salix psammophila.Their sequence analysis,phylogenetic tree and prediction of protein structure are finished by bioinformatics software.The results showed that the lengths of SpsLAZY1a and b were 1 143 bp and 1 161 bp,encoding 386 aa and 380 aa,respectively.Bioinformatics analysis showed that both SpsLAZY1a and b were hydrophilic proteins without signal peptide region.There existed typically conserved motifs of IGT gene family and C-terminal EAR structure of LAZY1 in their amino acid sequences.The 18 LAZY1 proteins were divided into two categories by phylogenetic tree in which SpsLAZY1a and b belonged to the first class (dicotyledon) and had high similarity as other plants.Amino acid homologous structure comparison showed that there were some similar homologous regions,but also some differences.Some regions existed in dicotyledon,but not in monocotyledon,and the same in herbaceous plant,but not in woody plants,LAZY1 gene duplication occurred during Salix evolution.SpsLAZY1a and b which belonged to the IGT gene family were cloned successfully from Salix psammophila and LAZY gene phylogenetic relationships were revealed initially in this research.
