摘要:
 试验从葡萄风信子“亚美尼亚”中利用RACE技术克隆了一个MYB类转录因子,命名为MaMYB2,Genebank登陆号为KJ744038。克隆到的MaMYB2全长711 bp,开放阅读框为600 bp,编码199个氨基酸,推测的蛋白分子量约为22.34 ku,理论等电点为5.46,保守域具有DNA结合位点;与矮牵牛(AAF66730.1)关系最近;核定位试验表明MaMYB2蛋白定位在细胞核中;实时定量PCR分析了MaMYB2在葡萄风信子“亚美尼亚”不同组织部位中的表达模式,结果不同组织中均有表达,在花中表达量最高,且随着花色变深表达量逐渐升高。结果表明,MaMYB2有可能参与了葡萄风信子中花青素积累的转录调控。
Abstract:
 In this study, we isolated a novel MYB transcription factor from Grape Hyacinths, named MaMYB2  and then analyzed its biological information and expression pattern. The ORF of MaMYB2 was 600 bp and encoding 199 amino acids. Sequences analysis showed that the deduced protein molecular weight of the gene was 22.34 ku. MaMYB2 protein was localized in the nucleus when it was transiently expressed in onion epidermal cells. Real-time quantitative PCR analysis revealed that MaMYB2 transcription increased in flowers compared with other organs. The gene expression level in flower tissues increased significantly during flower development, which is consistent with the blooming progress.
