摘要:
以文冠果成熟种子的子叶为外植体,优化培养基配方,诱导收集胚性愈伤组织,建立了高效的同步胚液体培养扩增体系,同步胚转移到固体培养基上,实现了植株的高效再生,为文冠果转基因平台的建立打下了基础。结果表明,文冠果子叶外植体胚性愈伤组织诱导优化培养基为附加2,4D 2.0 mg·L-1、NAA 1.0 mg·L-1、6BA 1.0 mg·L-1和3%的蔗糖的MS固体培养基,可产生大量胚性愈伤组织;胚性细胞系经继代和初步扩增后,在附加2,4D 0.5 mg·L-1、NAA 0.5 mg·L-1、6BA 0.5 mg·L-1和2%的蔗糖的B5液体培养基中增殖产生大量的同步球形胚;球形胚转入NAA 0.25 mg·L-1、6BA 0.5 mg·L-1的B5固体培养基上,经心形胚、鱼雷形胚和子叶形胚,再生成植株。
Abstract:
The cotyledons of Xanthoceras sorbifolia were selected as explants to induce somatic callus, with an optimized protocol, somatic callus were selected and subcultured to establish a high frequency proliferation system of synchronical somatic embryo in liquid medium. When the liquid cultured embryos were transferred on solid germination medium, plantlets were regenerated, which lays a platform for the future gene transformation of X. sorbifolia. The optimized medium as follows. Induction of somatic callus: MS medium supplemented with 2.0 mg·L-12,4D, 1.0 mg·L-1NAA, 1.0 mg·L-16BA, 3% sucrose and 0.66% agar; Synchronical globular somatic embryo liquid culture medium: B5 with 0.5 mg·L-1 2,4D, 0.5 mg·L-1 NAA, 0.5 mg·L-1 6BA and 2% sucrose; Somatic embryos development and germination medium: solid B5 with 0.25 mg·L-1 NAA and 0.5 mg·L-1 6BA. Heartshaped, torpedoshaped, and cotyledonshaped embryos were observed in germination process.
