摘要:
为探讨天女木兰组织培养中无菌外植体的有效获得,将天女木兰外植体分为种子和其他3种(侧芽、带侧芽的茎段、顶芽)外植体进行研究。天女木兰种子分为3种不同类型消毒,进行材料污染率、萌发率及成苗率的统计。其他3种外植体(侧芽、带侧芽的茎段、顶芽)采用L9(34)正交设计,统计无菌率。结果表明:天女木兰种子消毒时间为4mi n 的Ⅲ型试材为最适外植体,成苗率可达70%。采用正交设计的3种外植体的最优水平组合为A1B2C3,即外植体为侧芽,消毒剂为0.1%Hg Cl 2,消毒时间为12mi n。
Abstract:
To get nontoxi c explant fro m Magnoli a si ebol di i ti ss ue cult ure effectly ,t he explant of M.si eboldi i was di vi ded i nto seed and ot her t hree explants li ke s prout ,ste m of s prout and ter mi nal bud for researchi ng deeply .The arti cle takes 3ki nds of sterili zhi ng to M.si ebol di i to stati sti cs t he conta mi nati on rate ,ger mi nati on rate and seedli ng rate .Ot her explants (s prout ,ste m of s prout and ter mi nal bud)used L9(34)ort hogonal desi gn to mears ure pat hogen—free rate .The res ults s ho wed t hat t he best explant was Ⅲseed of M.si ebol di i ,its sterili zhi ng ti mes was 4mi n and seedli ng rate was hi gh to 70%.The best co mpose i n 3 types of explant usi ng ort hogonal desi gn was A1B2C3:explant was s prout ,di si nfectant was 0.1%Hg Cl 2,sterili zhi ng ti mes was 12mi n .
