摘要:
以杜仲优良品种的幼叶为材料提取总DNA,采用单因素梯度试验筛选法进行了杜仲RAPD反应的优化.确定的优化反应体系为:25 μL反应体积中,10×buffer 2.5 μL,TAqDNA聚合酶1 U,随机引物(5 μmol/L)2.0 μL,MgCl2(25 mmol/L)2.0 μL,dNTP(2.5 mmol/L)2.0 μL,模板2 mg/L,补ddH2O至25 μL.扩增程序为:94 ℃预变性5 min,94 ℃变性30 s,37 ℃退火30 s,72 ℃延伸90 s,35个循环,72 ℃延伸10 min,4 ℃保持产物.在此条件下得到的RAPD图谱清晰、稳定,为进一步利用该技术开展杜仲有关分子生物学研究提供了技术参考.
