摘要:
为了找出能够克服或降低高山杜鹃试管苗玻璃化的最适培养条件,本文采用单因子实验设计,以高山杜鹃茎尖作为外植体,Read为基本培养基,研究了培养基中添加物6-BA、蔗糖、琼脂、活性炭的浓度及培养温度等因子对高山杜鹃试管苗生长的影响.结果表明,6-BA、蔗糖、琼脂、活性炭的浓度及培养温度均对试管苗玻璃化率有显著影响.附加了1.5~2.0 mg/L 6-BA,3﹪~4﹪蔗糖,0.6﹪琼脂,0.3﹪活性炭的Read培养基和25℃的培养温度能有效得降低高山杜鹃试管苗玻璃化率,而且增殖系数较高、有效苗生长较快.
