摘要:
 以花椒试管苗叶片为试验材料,研究了不同激素浓度对花椒愈伤组织继代培养的影响,并以多次继代得到的疏松、活性高的愈伤组织建立花椒细胞悬浮系。结果表明:花椒愈伤组织最适继代培养基为MS+2,4-D 1.0 mg·L-1+6-BA 0.5 mg·L-1 +蔗糖 30 g·L-1 +琼脂 6 g·L-1;花椒细胞悬浮培养的最佳培养基和培养条件为:MS+2,4-D 1.0 mg·L-1+6-BA 1.0 mg·L-1+葡萄糖30 g·L-1,接种量0.05 g·mL-1,摇床转速110 r·min-1,25 ℃暗培养,该条件下悬浮细胞生长良好,最大生长量为4.52 g;花椒悬浮细胞生长呈“S”型曲线,最适继代周期为16 d。
Abstract:
 Plantlet leaves of Zanthoxylum bungeanum were taken as experimental materials for studying the effects of different concentrations of hormone on callus induction and subculture.The crisp callus was selected to establish cell suspension culture.The results showed that the best callus subculture medium was MS+2,4-D 1.0 mg·L-1+6-BA 0.5 mg·L-1+sucrose 30 g·L-1+agar 6 g·L-1.The best suspension culture medium for Z.bungeanum cells was MS+2,4-D 1.0 mg·L-1+6-BA 1.0 mg·L-1+glucose 30 g·L-1.The best culturing conditions were:inoculum size:0.05 g·mL-1,gyratory shaker rotation speed:110 r·min-1,temperature:25 ℃ in dark environment.Under these conditions suspended cells grew well and the maximum fresh weight of cells was 4.52 g.Suspended cells growth of Z.bungeanum exhibited “S-type” curve and optimal subculture cycle was 16 days.
