摘要:
以柴松不定芽为外植体,在保证基本培养条件不变的情况下,采用完全随机区组设计,研究培养基种类、活性炭浓度、蔗糖浓度、转接周期等对不定芽伸长生长的影响。结果表明:适宜于柴松不定芽伸长生长的基本培养基为1/2MS,不定芽生长健壮,苗高均值达到最大,为1.86 cm;活性炭浓度为0.1%时,有利于丛生芽苗的伸长生长,苗高均值达到1.48 cm;当蔗糖浓度为2.0%时,苗高均值最大,为1.28 cm;当培养周期为20 d时,有效地改善了芽苗枯死现象,培养周期延长至40 d时,芽苗枯死率明显升高。柴松不定芽生长与伸长的适宜培养基1/2MS+0.1 mg·L~(-1)AC+2.0mg·L~(-1)蔗糖。
