摘要:
 以毛竹根系作为试验材料,经过4种施肥处理(45%矿渣肥、39%矿渣肥、毛竹专用肥和对照),利用高效液相色谱法建立并优化了毛竹根系中有机酸的提取和测定方法,并测量了不同施肥处理下不同年龄毛竹根系中有机酸种类以及含量。结果表明,运用此方法样品制备简便,分离效果和方法的线性范围都能满足毛竹根系中有机酸定性和定量分析的要求。根据分析物的保留时间对样品色谱峰定性分析发现,不同施肥处理下各年龄毛竹根系中可以检出4种有机酸分别为苹果酸、草酸、柠檬酸和丁二酸。通过峰面积计算对有机酸定量分析发现:苹果酸含量总体表现为45%矿渣肥>对照>39%矿渣肥>专用肥;丁二酸含量总体表现为45%矿渣肥> 39%矿渣肥>专用肥>对照;而含量变化不大的草酸和柠檬酸均表现为45%矿渣肥>专用肥>39%矿渣肥>对照。在不同年龄毛竹根系有机酸含量的比较中发现,整体毛竹根系中有机酸含量随年龄的增长呈现出“大小年”的现象。
Abstract:
Bamboo roots were subjected to four fertilization treatments (45% slag fertilizer, 39% slag fertilizer, bamboo specialized fertilizer and control). The organic acids exudated from the roots were determined by high performance liquid chromatography (HPLC) method. Conditions for HPLC analysis were established. It was found that the method established was simple in sample preparation, the separation results and linear range could satisfy in qualitative and quantitative analysis of requirements. Four organic acids, such as malic acid, oxalic acid, citric acid and succinic acid were detected. The malic acid contents in the roots of 4 treatments were in an order of 45% slag fertilizer> control> 39% slag fertilizer> special fertilizer; the order of succinic acid content: 45% slag fertilizer> 39% slag fertilizer> special fertilizer> control; and oxalic acid and citric acid were 45% slag fertilizer> special fertilizer> 39% slag fertilizer> control. We also found that in the comparison of bamboo roots with different ages, the contents of organic acids demonstrated annul differences.
