摘要:
 为深入了解当前文冠果栽培群体和野生群体共存格局下的遗传多样性及其引种迁移格局变化,基于叶绿体测序分析数据,采用变异检测、多态性分析、单倍型分析、亲缘关系分析、Structure分析和主成分分析等方法,对9个野生群体和7个栽培群体共172个文冠果优树进行遗传多样性及其遗传结构研究。研究结果,为成功测序组装172个文冠果优树叶绿体基因组,均为典型的四分结构,基因组长度略小于参考叶绿体基因组,基因类别及数量与参考叶绿体基因组相当;共计检测到6类突变、52 460个SNP位点;其遗传多态性处于中低水平,各群体之间的遗传分化水平相对较低;多种分析方法得出的群体结构与单倍型分析结果基本一致,将16个群体分为4个组,根据地形位置可以划分为西部、中部、东北部和东南部。总体来说野生资源多样性较高,而栽培资源多样性较低。在当前野生群体与栽培群体共同存在的格局下,应优先保护野生群体,重点保护部分优株群体,可促进文冠果种质资源的有效保护与高效利用。
Abstract:
 In order to deeply understand the genetic diversity and the change of the introduction and migration patterns under the coexistence pattern of cultivated population and wild population of Xanthoceras sorbifolium,based on the chloroplast sequencing data,the genetic diversity and genetic structure of 172 X.sorbifolium superior trees from 9 wild populations and 7 cultivated populations were studied by using the methods of variation detection,polymorphism analysis,haplotype analysis,kinship analysis,structure analysis and principal component analysis.Totally,172 chloroplast genomes of X.sorbifolium superior trees were successfully sequenced and assembled,all of which were of typical tetrad structure.The length of genes was slightly smaller than that of the reference chloroplast genome,and the category and number of genes were equivalent to that of the reference chloroplast genome.A total of 460 SNP mutations were detected with the middle and low level genetic polymorphism,and the level of genetic differentiation among populations was relatively low.The population structure obtained by various analysis methods was basically consistent with the results of haplotype analysis.Sixteen populations were divided into four groups,which were divided into west,middle,northeast and southeast populations according to the topographic location.In general,the diversity of wild resources was high,while the diversity of cultivated resources was low.In the current pattern of the coexistence of wild population and cultivated population,we should give priority to the protection of wild population and focus on the protection of some excellent plant populations to promote the effective protection and efficient utilization of X.sorbifolium germplasm resources.
