摘要:
 建立利用高效液相法(HPLC)同时测定杜仲叶中绿原酸和芦丁的方法。色谱柱为Aglient ZORBAX SB-C18(4.6 mm×250 mm,5 μm),流动相组成为甲醇-乙酸-水(体积比为50∶1∶49),检验波长为340 nm,流量为1 mL/min,检测温度为32℃。在该色谱条件下,绿原酸和芦丁分别在0.060 0~2.000 0 μg/mL和0.001 8~0.090 0 μg/mL范围内线性关系良好,相关系数分别为0.999 7和0.999 3。利用此方法对杜仲杂交后代中41株生长性状表现优良的个体进行了叶片中绿原酸和芦丁含量的测定,为杜仲优良无性系的选择提供依据。
Abstract:
 A convenient high performance liquid chromatographic (HPLC) method has been established for the simultaneous determination of chlonogenic acid and rutin in the leaves of Eucommia ulmoides. Two components were saperated by the column Aglient ZORBAX SB-C18 (4.6 mm×250 mm, 5 μm) with a mobile phase of methanol-acetic acid-water (50∶1∶49, V/V) at a flow rate of 1 mL/min, detection wavelength of 240 nm and column temperature of 32℃. The chlonogenic and rutine each showed good liner relationship in a range of 0.060 0 μg/mL~2.000 0 μg/mL and 0.001 8 μg/mL~0.090 0 μg/mL, respectively, with the correlation cofficients of 0.999 7 and 0.999 3. The two components in the leaves of 41 fine individual clones were determined using the method above to provide basis for selection of superior clones.
