摘要:
 利用壳聚糖作为激发子,研究其对低温胁迫下核桃枝条相关酶活性与蛋白含量的影响,探讨低温胁迫条件下壳聚糖对核桃枝条抗冻反应的激发作用。通过对离体水培的香玲核桃枝条喷施1‰的壳聚糖,研究其在4℃低温胁迫6、12、24、36、48、72、96、120 h后相关酶活性、过氧化物与抗冻蛋白含量,分析低温胁迫后壳聚糖对树体抗冻相关酶活性和蛋白含量的激发作用。结果表明:在-2℃的低温胁迫下,施用壳聚糖后,核桃枝条内SOD、CAT的活性显著高于未施用的低温胁迫对照,但POD活性与壳聚糖施用未呈正相关性;同时低温胁迫组中施用壳聚糖后的MDA含量比未施用壳聚糖显著下降,而脯氨酸的含量有显著增加。壳聚糖能够显著提高低温胁迫下核桃抗氧化酶活性和脯氨酸含量,表明壳聚糖能够作为激发子增强核桃树体的抗冻反应。
Abstract:
 The effect of chitosan as an elicitor on metabolism of cold tolerance was studied by analyzing the activities of related enzymes and soluble protein content of walnut trees under the cold stress.The walnut branches from walnut breed “Xiangling” were treated by 0.1‰ chitosan under the stress of 4℃.The antioxidant enzyme activities and the contents of MDA,soluble proteins and proline were measured at 6,12,24,36,48,72,96 and 120 hours after the low temperature treatment.The results showed that the antioxidant enzyme activities of SOD and CAT increased dramatically when the chitosan was used as the elicitor compared to the control when the branches under the stress of cold at -2℃.However,the POD activity was not correlated with the elicitation of chitosan.The MDA content significantly decreased while the proline percentage was higher than the control under cold stress.The results suggested that the increase in antioxidant enzyme activities and the proline content was induced by the chitosan under the cold stress,indicating the chitosan could be an elicitor to activate the cold tolerance metabolism walnut tree.
